Raman for Bioprocessing

Raman spectroscopy has been seen as spectroscopic tool used in bioprocessing to monitor and control cell metabolism to optimize quality and yield of titer. This is due to Raman’s insensitivity to water and ability to analyze samples without manipulation. However there are certain challenges for Raman when analyzing a complex and dynamic sample such as a cell culture. This article will describe the different challenges in analyzing in-line Raman data and how to mitigate changes in samples that include changes in fluorescence background, presence of cells/debris in samples, and spectral interference. Mr. Justin Moretto's Abstract Kaiser Optical Systems, Inc. has been at the leading edge of in situ PAT in upstream bioprocessing for over 10 years. Kaiser Raman technology is robust, scaleable, and transferable. During process development, Raman enables adoption of QbD principles to define manufacturing design spaces, optimize process conditions, and demonstrate process robustness and quality. In production, improvements to product quality and yield have been demonstrated by employing Raman-based control strategies. In recent years, Kaiser Raman successes in upstream bioprocessing have expanded into downstream and cell / gene therapy applications. An overview of biopharma PAT milestones and Kaiser Raman technology advances will be presented. |
